The main objective of the game is that the player can choose one of several interconnected trucks and drive across the virtual map of Europe, collect cargo from different places and deliver them. As the game progresses, the player can buy more vehicles and warehouses, as well as hire other drivers to work on them.

Euro Truck Simulator 2 starts with the player choosing the location of his office in any of the cities on the virtual map. At first, the player can only complete the so-called Quick Quests - these tasks include delivering a hired driver while doing work in a delivery company, providing a truck and covering all expenses (fuel, road tolls, ferry crossings). As the player gradually earns money or can take out a bank loan, he can end up buying a truck and garage and start working in a higher paying job using his own truck, instead of being a hired driver with the equipment provided. The skills of the drivers that the player can hire over time increases with experience, and the player has the opportunity to create a large fleet of trucks and drivers, while expanding the business throughout Europe.

The base game includes 71 cities in twelve different countries, over twenty different types of cargo, and over fifteen fictional European companies.
